DESCRIPTION Gene Name Synonym FAS, APT1, FAS1, TNFRSF6, CD95 Species Mouse Accession P25446 Source HEK293 cells SPECIFICATIONS Amino acid sequence Gln22-Arg169 is expressed with a 10×His tag at the C terminus Molecular Weight 18 kDa(The protein migrates as 20-40 kDa under reducing condition (SDS-PAGE) due to glycosylation) Purity >95.0% as determined by SDS-PAGE Endotoxin <5 EU mg -1 Formulation Lyophilized from sterile PBS, pH 7.4.Normally 5%-8% trehalose and mannitol are added as protectants before lyophilization. Reconstitution Dissolved in sterile water, it is recommended to add 0.1% HSA or BSA for long-term preservation. STORAGE Stability and Storage 12 months from date of receipt at -20°C to -70°C. After reconstitution, it is stable at 4°C for 1 week or -20°C for almost 6 months (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage. BACKGROUND The Fas is also known as FAS receptor (FasR), apoptosis antigen 1 (APO-1 or APT), cluster of differentiation 95 (CD95) or tumor necrosis factor receptor superfamily member 6 (TNFRSF6). It is a prototype death receptor characterized by the presence of an 80 amino acid death domain in its cytoplasmic tail. This domain is essential for the recruitment of a number of signaling components upon activation by either agonistic anti-CD95 antibodies or cognate CD95 ligand that initiate apoptosis. The complex of proteins that forms upon triggering of CD95 is called the death-inducting signaling complex (DISC). The DISC consists of an adaptor protein and initiator caspases and is essential for induction of apoptosis. Changes in the expression of CD95 and/or its ligand CD95L are frequently found in human cancer. The downregulation or mutation of CD95 has been proposed as a mechanism by which cancer cells avoid destruction by the immune system through reduced apoptosis sensitivity. Thus, CD95 has therefore been viewed as a tumor suppressor. CD95 has been reported to be involved in the activation of NF-kappaB, MAPK3/ERK1, MAPK8/JNK, and the alternate pathways for CTL-mediated cytotoxicity. Accordingly, this protein is implicated in the pathogenesis of various malignancies and diseases of the immune system. Data SDS-PAGE Bioactivity Testing in progress Recombinant Mouse FAS Protein was determined by SDS-PAGE with Coomassie Blue, showing a band at 20-40 kDa. 仅供科研用途,不可用于临床诊断!
