BACKGROUND PD-L1, which we first identified as a homologue of B7 family molecules, is expressed on immune cells, i.e., dendritic cells (DCs) and B and T cells, and controls T-cell immune responses such as peripheral tolerance, termination of immune responses and immune exhaustion after prolonged exposure to an antigen stimulus. PD-L1 plays a crucial role in the disease progression of multiple myeloma. PD-L1 has been demonstrated to play a role in the regulation of immune responses and peripheral tolerance. PD-L1 can bind to PD-1 expressed on cytotoxic T lymphocytes (CTLs), resulting in the escape from immune surveillance by the induction of CTL apoptosis. PD-L1 is regarded as promising therapeutic target for human autoimmune disease and malignant cancers.
